So when the game asks us to "become human" what does that mean? In the case of Kara, it means she was empathetic of her ward Alice, and broke free of her coding as it was not allowing her to protect Alice. For Markus his moment of becoming human is caused by a personal injustice, where he is told not to defend himself against the barage of hits. He decides it is not fair to have zero say in what happens to him and decides the choices will be his to make from now on. When Connor becomes human, it is because he sees the humanity in the deviants he was assigned to hunt and comes to realize his assignment is against his new beliefs in androids as sentient beings. 

This idea of becoming human may be a surprise to see in modern video games, as it is quite a posthuman concept. It asks us to identify the process of becoming, instead of looking at exactly what our characters are in this moment or what they possibly will be. This process is empasized in Detroit Become Human to come from empathizing with androids as equal sentient beings, especially as shown through Kara and Alice's relationship. In the case of Markus we see a more freedom based idea of humanity,  where injustice and freedom are the basis of his change and his following story arc. It is also interesting that much of the choices to be made in the game come after the characters have decided to "become human", as Kara and Markus both begin the process in their second chapters.
