DeLou Burke was born on 24 March 1858 in Crawfordsville, Indiana. He attended Central Normal School in Danville, Indiana, and then taught school until 1888. He then went to Nashotah House Seminary, graduating in 1892 and being ordained by Bishop Nicholson at Milwaukee. is first assignment came at St. Mark's Church in Beaver Dam, Wisconsin. He then went to South Bend, where he took charge of both St. James Church and St. Paul's, Mishawaka. In 1898 he left to become record of St. James, Vincennes, and also accepted a post as chair of the Philosophy Department of Vincennes University. From here he moved to Topeka, Kansas, and became dean of Grace Cathedral. He died in Montgomery, Indiana, on 8 May 1908, and was buried in Riverview Cemetery, South Bend. He was unmarried.
